纤维粘连蛋白基因ED-B片段在涎腺肿瘤中的表达

     [摘要] 目的:研究纤维粘连蛋白基因ED-B片段在涎腺正常组织及其良、恶性肿瘤组织中的表达情况,探讨其与涎腺良、恶性肿瘤发生、发展的关系。方法:分别提取涎腺正常组织及其良、恶性肿瘤组织的总RNA,RT-PCR法扩增ED-B片段,重组于pGEM-T easy 载体, 酶切制备成模板,体外逆转录方法合成cRNA探针,通过RT-PCR和原位杂交方法分别检测ED-B mRNA的表达情况。结果:ED-B-在正常涎腺及其良、恶性肿瘤组织中均呈高表达;ED-B+在正常腺体组织中不表达,而在良性肿瘤中有表达,且在恶性肿瘤中其阳性表达程度增高。结论:随着涎腺向肿瘤、恶性肿瘤方向的转化,FN基因ED-B剪切片段的表达逐渐增强。

    The Expression of ED-B Isoform of Fibronectin gene in Oral salivary gland tumors.

    [Abstract]Objective: The aim of this study was to investigate the expression of ED-B isoform of fibronectin in normal, benign, and malignant tissues of salivary gland, and find the relation ship of ED-B and generation, development and matastasis of salivary gland tumors. Methods: Total RNA was extracted from normal, benign and malignant tissues, ED-B DNA bands were obtained by RT-PCR. The fragments of cDNA were cloned into the pGEM-T easy vector. The sense and antisense templets were made by digestion of the plasmid pGEM-T easy of ED-B with restriction enzymes. The cRNA probes were constructed from the templets by transcription in vitro. Expression of ED-A-, ED-B- and ED-B+ mRNA in different tissues were analyzed by in situ hybridization. Results: The expression of ED-B- was found in all the cases. The expression of ED-B+ had been found to be absent in normal tissues, and was much greater in malignant tissues than those in benign tissues. Conclusions: The expression of ED-B of fibronectin gene increased as with salivary tumors transforming to malignant. ......
