微量注射甲基软海绵酸对大鼠海马神经元放电的影响

     【摘要】 目的探寻新的建立老年痴呆模型的方法。方法大鼠海马背侧微量注射0.4 mmol/L 甲基软海绵酸(Okadaic acid,OA)1 μ1,每两天 1 次,连续 3 次。海马背侧首次微量注射20 d后,测试各组大鼠空间学习记忆能力;行为学检测结束后,在注射对侧海马观察各组神经元自发放电。结果海马背侧微量注射OA大鼠空间学习记忆能力明显降低,海马神经元放电频率明显减少,放电形式发生改变。结论OA对海马影响明显,提示此法可建立老年痴呆模型。

    The effect of the discharge of hippocampus neurons of rats by injection of okadaic acid

    【Abstract】Objective In order to explore a new way of establishing Alzheimer's animal model. MethodsOkadiac acid(0.4mmol/L1 μl) was injected into the right dorsal hippocampus,once every two days for three times.The place navigation and spatial probe ability in rats were assessed by Morris water maze on day 20 after the first injection.After the spatial probe test,the spontaneous discharge was observed in the left hippocampus. ResultsBy injecting okadaic acid into the dorsal hippocampus,the spatial learning and memory ability in rats was significantly impaired by okadaic acid,the discharge frequency of hippocampus neurons was signifcianly reduced,and the format of the discharge was also changed.ConclusionOkadaic acid affected the hippocampus significantly.It suggests that this method can be used to establish the experimental model of AD. ......
