一氧化氮对大鼠模拟缺血再灌注心肌细胞凋亡的影响

     【摘要】 目的 探讨NO对心肌缺血再灌注(I/R)细胞凋亡的影响。方法 将原代培养新生大鼠心肌细胞随机分为(1)正常对照组及I/R对照组、(2)NO供体L-精氨酸(L-Arg)组、(3)NOS抑制剂L-硝基-精氨酸(L-NNA)组。心肌细胞用模拟缺血液培养2h后,再用模拟再灌注液培养1h创建缺血再灌注模型,L-Arg组和L-NNA组于再灌注时分别给予L-Arg和L-NNA与心肌细胞共同孵育。以LDH的含量和心肌细胞凋亡百分率以及凋亡指数为损伤检测指标。结果 心肌细胞缺血再灌注后,细胞培养上清液中NO含量和LDH含量增加,TUNEL染色检测到大量阳性凋亡细胞;L-Arg组NO、LDH含量以及细胞凋亡率和凋亡指数均较对照组明显增加,而L-NNA组上述指标均显著降低。结论 NO可通过诱导心肌细胞凋亡而加重缺血再灌注损伤。

    The effect of nitric oxide on ischemia-reperfusion cadiocyte apoptosis of rats

    【Abstract】 Objective To obseve the effect of nitric oxide(NO )on ischemia-reperfusion(I/R) cadiocyte apoptosis.Methods Primary cultured cadiocytes of neonatal rats were random divided into (1)normal control group and I/R control group,(2)NO donor L-Arg group and,(3) nitric oxide synthase(NOS) inhibitor L-NNA group. Checking up the levels of NO and LDH in each group, and apoptosis was detected by TUNEL and flow cytometer.Results After I/R, levels of NO and LDH had been risen clearly, and the index and the percentage of apoptosis increased obviously too. In L-Arg group, the former index constantly risen,but in L-NNA group ,the former index obviously decreased,compared with control group.(both P<0.01).Conclusion NO may incresae rats cardiomyocytes I/R injury by induced cadiocytes apoptosis. ......
