摘要:目的 制备盐酸万乃洛韦微孔渗透泵片,考察该制剂的释药影响因素及释药机理。方法 利用正交设计优化处方,根据不同时间累积释放度考察药物的释放情况。结果 盐酸万乃洛韦微孔渗透泵控释片体外释药速度较市售片缓慢、平稳。结论 本制剂释药速率随渗透压差减小而成比例减小,遵从以渗透压差为释药动力的释药模式,12 h内呈现良好的零级释放特征。

Preparation of valaciclovir hydrochloride micro-porous osmotic pump tablets and study on the mechanism of drug release

Abstract:Objective To prepare oral micro-porous osmotic pump tablets and investigate the factors influencing the drug release and drug release mechanism..Methods The optimal formulation of micro-porous osmotic pump-controlled release tablets were screened by the orthogonal design. Results The valaciclovir hydrochloride micro-porous osmotic pump tablets were prepared, of which the release rate was slower than that of the commercial conventional tablets. Conclusions The release profile of this formulation fit to zero-order release mechanism during 12 h. The drug release rate decreased in proportion to the osmotic pressure difference, which proved its osmotic principle.
