砷对小鼠肾脏磷脂脂肪酸的影响及Vit E干预作用的研究

     摘要: 目的: 研究不同水平砷对小鼠肾脏组织磷脂脂肪酸组成的影响,同时观察维生素E(VitE)对砷毒作用的干预作用。 方法: 按两因素三水平析因实验设计将昆明种小鼠分为9组,复制亚慢性砷中毒动物模型,并进行Vit E干预研究。2个月后,采用薄层气相色谱法,研究砷对小鼠肾脏组织中磷脂脂肪酸组成的影响以及给予Vit E处理后的干预作用。 结果: 与正常对照组相比, 染毒组动物肾脏组织磷脂脂肪酸中饱和脂肪酸含量明显升高(P<0.05),多不饱和脂肪酸含量明显下降(P<0.05~0.01);给予Vit E处理后,动物肾脏组织磷脂脂肪酸中饱和脂肪酸含量明显降低(P<0.05~0.01),多不饱和脂肪酸含量明显升高(P<0.05~0.01)。 结论: 砷可在小鼠肾脏组织中蓄积造成磷脂脂肪酸组成发生改变,即多不饱和脂肪酸组成下降,饱和脂肪酸组成升高;VitE可拮抗砷对小鼠肾脏组织中磷脂脂肪酸组成的影响,使饱和脂肪酸和多不饱和脂肪酸组成趋于正常。

    Experimental study of arsenic and the antagonistic action of vitamin Eon fatty acid metabolism of phospholipid in the kidney of mice

    Abstract: Objective: To study the influence of arsenic and the antagonistic action of vitamin E on phospholipid fatty acid metabolism in the kidney of mice. Methods: Kunming mice were divided into nine groups according to the factorial experiment design of two factors and three levels. The mice of different groups were fed with both various amounts of arsenic in drinking water and vitamin E for two months to produce the animal model of subchronic arsenic poisoning. Phospholipid in the mice kidney was extracted by the BlighDyer method and fatty acid compositions of the phospholipid were assayed using gas chromatography. Results: (1) The fatty acid compositions of phospholipid in arsenic groups changed abnormally, which the proportion of unsaturated fatty acids reduced and the saturated fatty acids increased (P<0.05). While, in vitamin E (Vit E)+ Arsenic(As) groups, the proportion of unsaturated fatty acids increased and the saturated fatty acids reduced nearly to normal level (P<0.05). Conclusions: (1) The high dose of arsenic can be accumulated and it can cause the abnormal changes of the fatty acids of phospholipid in the kidney of mice, which might be the result from oxidative stress and the increased level of lipid peroxidation. (2) There is an interaction between arsenic and vitamin E. Vitamin E has an antagonistic action against the arsenism through protecting the normal compositions of fatty acids of phospholipid and mataining the balance of the lipid peroxidation and antilipid peroxidation system. ......
