酒精合并高脂饮食性脂肪肝大鼠模型的建立

    摘要: 目的:建立符合现代人饮食习惯的脂肪肝大鼠模型。方法:Wistar大鼠40只,随机分为4组,每组10只。正常组每天15 ml/kg生理盐水灌胃,模型1组、2组每天分别给予15 ml/kg 1号脂肪乳、15 ml/kg 2号脂肪乳灌胃,并均同时喂饲普通饲料,模型3组每天15 ml/kg高度白酒灌胃同时给予高脂饲料,连续4周,分别测定各组大鼠的肝指数、血液流变学指标、羟脯氨酸(HYP)、血清和肝组织中脂质含量,并进行肝脏病理检查。结果:各模型组大鼠肝脏病理检查结果均表现为广泛的脂肪变性;模型1组与模型2组肝指数、血清中总胆固醇(TC)、肝脏组织中甘油三酯(TG)、HYP含量均明显高于正常组(P均<0.01),而血清中TG、低切全血粘度、高切全血粘度、红细胞聚集指数均明显低于正常组(P均<0.01);模型3组HYP含量明显高于正常组(P<0.01) ,而血清TG、低切全血粘度、红细胞聚集指数均明显低于正常组(P均<0.01)。结论:建立了酒精合并高脂饮食性脂肪肝大鼠模型,为脂肪肝的实验研究提供了理想的动物模型。

    Establishment of rat fatty liver model about alcohol combine with high fat diet

    Abstract: Objective: To set up rat fatty liver model according with modern people eating habit. Methods: There are 40 rats which were divided 4 groups at random. The control group were intragastric infused of nomal saline 15 ml/kg everyday, the animal group 1 were intragastric infused of fat galacta 1 15 ml/kg everyday, the animal group 2 were intragastric infused of fat galacta 2 15 ml/kg everyday, the animal group 3 were intragastric infused of spirit 15ml/kg everyday and fed with high fat diet. All rats were sacrificed at the end of the fourth week, then hepatic index, hemorheological parameters, hydroxyproline(HYP), blood lipid, liver lipid, and liver pathology. Results: Wide hepatic steatosis was induced in all model groups. Liver index , serum total cholesterol(TC) , liver triglyceride(TG) , HYP were higher in the model group 1 and the model group 2 (P<0.01) than the control group; but serum triglycer (TG), blood viscosity (1s1, 200s1), erythrocyte aggregation index were lower in the model group 1 and the model group 2 (P<0.01) than the control group. HYP were higher in the model group 3 (P<0.01); but serum triglycer (TG), blood viscosity (1s1), erythrocyte aggregation index were lower in the model group 3 (P<0.01) than the control group. Conclusion: The rat fatty liver model about alcohol combine with high fat diet is set up, and providing ideal animal model for fat liver resecrch. ......
