抗肺纤Ⅰ和Ⅱ号中药对大鼠肺间质纤维化的干预作用

     【摘要】 目的 探讨中药对肺间质纤维化的预防和治疗作用。方法 Wistar大鼠160只,随机分为8组:(1)对照组[(1)组]:灌胃生理盐水(NaCl)2.25mg/(kg·d);(2)强的松组[(2)组]:灌胃强的松6mg/(kg·d);(3)预防Ⅰ组[(3)、(4)组]:灌胃抗肺纤Ⅰ号7.5g/(kg·d)、15g/(kg·d);(4)预防Ⅱ组[(5)、(6)组]:灌胃抗肺纤Ⅱ号7.5g/(kg·d)、15g/(kg·d);(5)治疗组[(7)、(8)组],分别灌胃抗肺纤Ⅰ和Ⅱ号15g/(kg·d)。(1)~(6)各组于气管内滴入博莱霉素(BLMA5) 5mg/kg,次日开始给药。(7)、(8)组于气管内滴注BLMA5,5mg/kg,15天后开始给药。各组动物分别于给药14天、28天随机处死10只,然后检测各项观察指标。结果 观察28天后,对照组:动物体重增加71%(P<0.05),肺胶原蛋白含量提高17.7%(P<0.05),过氧化脂质含量提高1.86倍(P<0.05),羟脯氨酸含量提高24.01%(P<0.05)。强的松组:动物体重减轻80%(P<0.01),肺胶原蛋白降低35.3%(P<0.01),羟脯氨酸含量降低44.2%(P<0.01)。预防组:动物体重分别增长55.8%(P<0.05)和47.0%(P<0.05),肺胶原蛋白含量降低36.2%(P<0.05)和26.5%(P<0.05),羟脯氨酸含量降低24.9%(P<0.05)和70.7%(P<0.05)。治疗组肺胶原蛋白降低13.0%(P<0.01)和31.0%(P<0.05),羟脯氨酸含量降低30.0%(P<0.05)和21.0%(P<0.05)。结论 抗肺纤Ⅰ和Ⅱ号均具有预防和治疗肺间质纤维化的作用。

    The inhibitory effect of antipulmonary fibrosis agents Ⅰ and Ⅱ on the rat model of blemycin-induced pulmonary fibrosis

    【Abstract】 Objective To study the effect of antipulmonary fibrosis agent Ⅰ and Ⅱ on the rat model of blemycin-induced pulmonary fibrosis.Methods Wistar rats induced by intratracheal instillation of blemycin were randomly divided into six groups with twenty each group.Then bronchus was recepted daily saline 2.25mg/kg(group A),prednisone 6mg/kg(group B),APFAⅠ 15g/kg(group C) and APFAⅠ 15g/kg(group D) from the next day respectively.The rats in group E and F received daily APFAⅠ 15g/kg and APFAⅠ 15g/kg respectively from the 15th day.Ten rats in each group were randomly sacrificed 14 and 28 days after intratracheal instillation.Then the rats weight collagen,hydroxyproline of the lung tissue were detected.Results Collagen of rat lung tissue in group B,C,D,E and F decreased 35.3%,36.2%,25.6%,13.0% and 31.0% (five of P<0.05,respectively),meanwhile hydroxyproline decreased 44.2%,24.9%,70.2%,30.0% and 21.0% (five of P<0.05),respectivety.Conclnsion The anti-pulmonary fibrosis agents Ⅰ and Ⅱ can prevent and treat blemycin-induced pulmonary fibrosis in rats. ......
