供体脾脏对大鼠移植胰腺细胞凋亡的影响

     摘要: 目的: 观察供体脾脏对胰腺移植大鼠免疫反应的影响。方法: 制作异基因大鼠胰、脾、十二指肠联合移植模型,观察供体胰腺的病理学改变,测定供体胰腺组织中细胞凋亡的变化、转化生长因子β1(TGFβ1)mRNA的表达及CD4+ T淋巴细胞和CD8+ T淋巴细胞的变化。结果: 胰、脾、十二指肠联合移植组:术后3 d胰腺小叶间质水肿,腺泡细胞和胰岛细胞轻度水肿;术后5 d炎细胞浸润,腺泡细胞灶状坏死;术后7 d腺泡细胞和胰岛细胞部分坏死、自溶。胰、十二指肠移植组:术后3 d胰腺腺泡细胞片状坏死;术后5 d胰腺小叶和部分胰岛细胞坏死;术后7 d腺泡细胞大片坏死,胰岛结构消失。胰、脾、十二指肠联合移植组供体胰腺组织中凋亡细胞数和TGFβ1 mRNA的表达于术后各时点均高于胰、十二指肠移植组(P<0.001);其CD4+和CD8+T细胞数高于正常对照组,但低于胰、十二指肠移植组(P<0.001)。结论: 细胞凋亡和TGFβ可减轻受体大鼠对供体胰腺的免疫排斥反应;调高供体胰腺组织TGFβmRNA的表达是供体脾脏发挥免疫抑制作用的机制之一。

    Donor spleen influence the apoptosis in the transplanted pancreas in rat

    Abstract: Objective: To observe the influence of the donor spleen on the immunity reaction at the transplanted pancreas. Methods: The rat model of pancreas allotransplanted combining with spleen and duodenum were Manufactured. Pathology change of donor pancreas was observed. The change of apoptosis, the expression of the transforming growth factor β1 ( TGFβ1 ) mRNA and the variation of CD4+ T lymphocyte and CD8+ T lymphocyte in donor pancreatic tissue were measured. Results: In the first group of which pancreas was allotransplanted combining with spleen and duodenum: dropsy of stroma between pancreatic folia, light dropsy of gland alveolus cells and pancreatic islet cells were observed at 3 d post operation; Infiltration of inflammatory cells in pancreatic tissue and focus necrosis of the gland alveolus cells occurred at 5 d post operation; Necrosis and dissolve at part of gland alveolus cells and pancreatic islets were observed at 7 d after operation. In the second group of which pancreas allogenic graft was made combining with duodenum: schistic necrosis of pancreatic gland alveolus at 3 d post operation, necrosis of pancreatic folia and pancreatic islet at 5 d post operation, widespread necrosis and disappear of structure at 7 d post operation were observed. The number of apoptosis cells and the expression of TGFβ1mRNA in donor pancreas in the first group was higher at every time point than that in the second group. The number of CD4+ and CD8+T lymphocyte in donor pancreas was higher than that of control group, but lower than that of the second group. Conclusion: Apoptosis and TGFβ can reduce the immunity rejection that come from recipient and aim to donor pancreas. Upregulation of expression of TGFβmRNA in donor pancreas is one of the mechanisms through which donor spleen perform its immunosuppressive function. ......
