博莱霉素致大鼠肺纤维化模型的建立方法及比较

     摘要: 目的: 采用不同方法诱导博莱霉素(BLM)致大鼠肺纤维化的模型,探讨符合人类肺纤维化病程的动物模型。方法: 分别经气管、腹腔给予BLM致大鼠肺纤维化模型,于第1、14、28天处死,取肺组织制备病理切片,行HE染色后评价两种动物模型各时间段肺组织形态学变化,测量肺泡壁和胸膜厚度;测定肺组织匀浆中羟脯氨酸(HYP)含量,对两种模型进行评价。结果: (1)气管内、腹腔内给药均可建立肺纤维化动物模型,但时间长短不一;(2)气管内给药大鼠肺内病灶分布以末梢细支气管为主,早期的肺内炎症变化较重,而腹腔内给药大鼠病灶分布以胸膜下、肺组织内为主,炎症反应相对较轻;(3)腹腔内给药大鼠第28天肺组织匀浆HYP含量明显高于气管内给药(P<0.05)。结论: 腹腔内给药建立的大鼠肺纤维化模型可能较近似于人类特发性肺纤维化的病程及病理改变。

    Comparation of the model of bleomycininduced pulmonary fibrosis in rats with different methods

    Abstract: Objective: To provide a better model matching with human pulmonary fibrosis by studying a model of pulmonary fibrosis in rats. Methods: The Wistar rats were administerted with intraperitoneal or tracheal injection of bleomycin (BLM), and were sacrificed after treatment on days 1, 14 and 28, respectively, and their lungs were harvested for histological studies. After hematoxylin and eosin staining, histological changes were evaluated on 1, 14, 28 days. In addition, width of alveolar septa and pleura depth, and hydroxyproline in lung issues hemogenate were measured. Results: (1)Both of the methods could induce pulmonary fibrosis, but time was different;(2)BLM was mainly distributed at telebronchi by tracheal injection, while BLM was mainly distributed under pleura by intraperitoneal injection. The degree of alveolitis of the former was higher than that of the latter(P<0.05). (3)The hydroxyproline content in lung issues hemogenate of the former was higher than that of the latter at 28th day(P<0.05). Conclusion: The process and histological changes of the model for pulmonary fibrosis in rats established by intraperitoneal administertion was similar to that of human IPF, possiblely. ......
