     【摘要】 目的 建立一种简便、灵敏和快速分离测定鼠血清中腐胺、精脒和精胺等多胺含量的高效液相色谱-质谱联用方法。方法 采用苯甲酰氯衍生,选择性离子监测(SIM),方法的检测时间在10min以内。结果 腐胺、精脒、精胺定量检测限均达100pg/ml,日间和日内的测定精密度分别为2.4%~4.1%和1.9%~3.6%,回收率为90.8%~94.1%。结论 将该法用于胰腺癌裸鼠血清中多胺的测定,建立了一种胰腺癌药物筛选及其疗效评价的新方法。

    【Abstract】 Objective A simple, sensitive and fast method used for determining putrescine, spermidine and spermine in mouse serum using LC/ESI/MS has been proposed. Methods Benzoyl chloride was employed to derive polyamines to get better separation and detection limit. An ESI interface and the selected ion monitoring (SIM) mode were used. Results Satisfactory separation was achieved on an Agilent ODS-C18column by using the gradient elution with mobile phase consisting of methanol and water. The LC/ESI/MS analysis process took less than 10 minutes. The limits of quantification were all found up to 100 pg/ml for putrescine ......
