番泻叶提取物诱导小鼠结肠组织中蛋白质的差异表达

    摘 要:目的 建立和优化蛋白质分子差异展示的双向聚丙烯酰胺凝胶电泳技术,初步观察番泻叶提取物诱导小鼠结肠组织中蛋白质的差异表达,为进一步筛选其中介导番泻叶生物作用的蛋白质分子奠定基础. 方法 应用番泻叶提取物灌胃制备小鼠慢性胃肠运动增强模型,提取其结肠组织中总蛋白质,优化蛋白质分子差异展示的双向聚丙烯酰胺凝胶电泳技术,并用其分析模型动物结肠组织中蛋白质,双向电泳凝胶银染法显示其蛋白质的差异表达. 结果 建立了小鼠慢性胃肠运动增强模型,优化了双向聚丙烯酰胺凝胶电技术并用其展示出模型动物结肠组织中有差异蛋白质的表达. 结论 番泻叶提取物可以诱导小鼠结肠组织中蛋白质差异表达,这些差异蛋白质可能介导了番泻叶的生物作用.

    sional(2-D)polyacrylamide gel electrophoresis(PAGE)for investigating the different expression of colonic proteins in BALB/c mice induced by extract of senna,and to lay the foundation for screening and characterization of biological molecules mediating the effect of senna from the gastroin-testinal tract.METHODS The model of chronic gastroin-testinal motility hyperfunction in mice was established by in-tragastric administration of senna extraction.The proteins of colon was extracted from control and model mice.The2-D PAGE was optimized for analyzing the different expression of proteins.The2-D PAGE gels were stained with silver stain┐ing.RESULTS An animal model of chronic gastrointestinal motility hyperfunction was established in mice.Optimized2-D PAGE displayed the different expressional proteins in gas-trointestinal tract of model mice.CONCLUSION The dif-ferent expression of colonic proteins can be induced by extract of senna in BALB/c mice.These proteins maybe mediate the effect of senna in gastrointestinal tract. ......
