葛根素对肾缺血/再灌注时肾小管上皮细胞cFos蛋白表达的影响

     摘要:目的 从细胞凋亡角度研究葛根素治疗肾缺血/再灌注损伤的机制。方法 用肾动脉钳夹法建立大鼠肾缺血再灌注损伤模型,治疗组于肾缺血/再灌注前应用葛根素30mg/kg静脉注射;采用原位末端标记法、免疫组织化学法检测缺血再灌注不同时间点肾小管上皮细胞凋亡及cFos蛋白表达的变化。结果 葛根素能减少细胞凋亡,下调cFos蛋白的表达。结论 葛根素对急性肾缺血/再灌注损伤具有保护作用,其机制与下调凋亡相关基因cFos的表达有关。

    Effects of puerarin injection on cFos protein of renal tubule cells

    ABSTRACT: Objective To investigate the role of puerarin on acute renal ischemiareperfusion injury and its mechanism. Methods Acute renal ischemiareperfusion iniury rat model was established by placing the atraumatic vascular clamp in renal pedical for 30 minutes. Rats in puerarin group were given puerarin injection 30mg/kg before ischemia. Immunohistochemical methods were used to detect the changes of expression of cFos proteins; pathomorphological changes of renal ischemiareperfusion injury were alse observed, and the occurrence of apoptosis was detected by TUNEL method. Results The pathological changes mainly occurred in the tubular epithelial cells of rat kidneys.The apoptosis index increased in the kidneys of ischemiareperfusion group and puerarin group,and peaked at 48 h of reperfusion(P<0.01). cFos protein mainly expressed in renal distal convoluted tubules, but the expression of cFos in puerarin group was still less than that in the ischemiareperfusion group. In control group, apoptosis of renal tubule cells and cFos protein expression was little. Conclusion Chinese herb puerarin can protect kidney against ischemia after acute renal ischemiareperfusion injury, which may be associated with decreased cFos protein expression in rats. ......
