脑缺血性损伤后的皮层突触再生与运动功能恢复

    摘 要:目的 研究大鼠缺血后,损伤区域及相关部位神经再塑与运动行为恢复之间的关系. 方法 线栓法建立大鼠的大脑中动脉缺血模型,术后7,14,21,28d,采用免疫组织化学方法观察突触素(SYN)及突触后致密物质-95(PSD-95)在大鼠皮层缺血灶的内侧、外侧、对侧的表达,触觉刺激试验作行为测试. 结果 SYN及PSD-95的免疫反应产物于术后7d在3个观察部位都显著减少,在14d以后明显增加,与此相对应的肢体活动在21d明显恢复. 结论 缺血损伤后的大鼠皮层梗塞区的内侧、外侧、对侧的神经再塑、突触重建与其肢体的功能恢复有着明确的相关关系.

    Abstract:AIM To evaluate the distribution and density of expression of proteins known to be involved in synaptognesis and to observe the motor function of forelimbs after focal is-chemia in rats.METHODS Unilateral cerebral ischemia was induced in rats by the middle cerebral artery occlusion(MCAO).Synaptophysin and PSD-95were examined by im-munohistochemical techniques.Forelimbs function was eval-uated with tacticl stimulation test.RESULTS Both synapto-physin and PSD-95demonstrated statistically significant in-crease in the density of immunoreaction in the neocortical re-gions medial and lateral to the infarction as well as in the con-tralateral neocortex at14th day compared with sham con-trols.Meanwhile ......
