    Abstract

    AIM: To investigate the effect of infusing ex vivo isolated CD4+CD25+ regulatory T cells on the rejection model of DA-Lewis rat liver transplantation.

    METHODS:CD4+CD25+ regulatory T cells were isolated from Lewis (RT11) rat splenocytes and peripheral lymph nodes by magnetic cell sorting system (negative selection and positive selection), and co-cultured with mitomycin-C (MMC) treated dark agouti (RT1a, DA) rat splenocytes for 3 d. The suppressive efficiency of fresh or ex vivo stimulated CD4+CD25+ regulatory T cells on the proliferation of CD4+CD25- T cells was evaluated by cell proliferation assay.
