     Inhibitive effects of EB1089 on human hepatocellular carcinoma cells HepG2

    【Abstract】 AIM: To study the inhibition of EB1089 on human hepatocellular carcinoma cells HepG2 in vitro. METHODS: HepG2 cells were exposed to EB1089 at 1, 10, 100 nmol/L for 2, 4, 6 and 8 d, respectively. The inhibitory rate of cells was determined by cell counting and MTT assay and the cell growth curve was made. The cell cycle was monitored by flow cytometry analysis (FCM). RESULTS: EB1089 at 1-100 nmol/L inhibited the growth and proliferation of HepG2 cells. The inhibitory rates of the cells increased from 16.9% to 74.3% in comparison with those of the control (P<0.01), and the cell growth curve changed accordingly. After 4 d of exposure, 50% of the inhibitory concentration (IC50) of EB1089 was 8.2 nmol/L. FCM showed that the cell cycle of HepG2 cells was blocked in the phase transition from G0/G1 to G2. When the cells were treated with 10 nmol/L EB1089 for 4 d, FCM showed that apoptotic rate of the cells increased by 21.4 % (P<0.01). CONCLUSION: EB1089 can significantly inhibit the growth and proliferation of hepatocellular carcinoma cells HepG2. The block in the phase transition from G0/G1 to S may account for this inhibitory effect, and the induction of apoptosis may be responsible for the inhibition. ......
