     Effects of Glivec and hexamethylenebisacetamide on proliferation and apoptosis of K562 cells

    【Abstract】 AIM: To explore how Glivec and HMBA affect the cell growth, apoptosis, cell cycle and cyclinprotein expression of K562 cells. METHODS: After K562 cell line was cultured and Glivec and/or HMBA were added, the cell growth index and surviving rate were determined with MTT assay, the cell cycle and apoptosis were examined by flow cytometry and electron microscopy, the expression of bcrabl gene was examined by RTPCR, and the expressions of cyclinD1, cyclinE, cyclinA, CDK4 and cmyc protein were examined by Western blot. RESULTS: A combination of two drugs apparently reduced the surviving rate and slightly declined the percent of apoptosis. K562 cells were arrested in G0/G1 phase, and the expressions of cyclinD1, cyclinE, cyclinA, CDK4 and cmyc protein were downregulated. CONCLUSION: A combination of two drugs can effectively inhibit the growth and slightly inhibit the apoptosis of K562 cells. The action of drugs bears on cell cycle arrest, and downregulation of cyclin protein and cell apoptosis. ......
