     Construction of pIRES2EGFPhVEGF165 eukaryotic expression plasmid and its expression in cardiac myocytes in vitro and vivo

    【Abstract】 AIM: To construct a eukaryotic expression plasmid pIRES2EGFPhVEGF165 and to transfect it into cardiac myocytes in vitro and vivo. METHODS: The hVEGF165 DNA fragment was inserted into a green fluorescent protein fusion vector, which was then transfected into cardiac myocytes in vitro and vivo. RESULTS: A recombinant eukaryotic expression plasmid pIRES2EGFPhVEGF165 was constructed and was successfully transfected into cardiac myocytes. The expression of VEGF165 was confirmed by fluoroscopy and immunohistochemical staining. CONCLUSION: VEGF can be expressed stably in cardiac myocytes, which might be useful for gene therapy of myocardial ischemia. ......
