    Pulmonary CGRP and 5HT immunoreactive cells in developing rat lungs

    【Abstract】 AIM: The paper is to explore the development of calcitonin generelated peptide (CGRP) and serotonin (5HT) immunoreactive cells in lung and reveal the mechanism of the endocrine system. METHODS: The lung tissue was studied with immunohistochemitry and laser scanning confocal microscopy. The results were dealed with computer image analysis. RESULTS: Cells containing CGRP and 5HT firstly appeared in the 16day fetuses, and increased in number as gestation progressed and reached its peak at 20day fetuses. The number of CGRP and 5HT positive cells declined in postnatal rat lungs(NE/per 10 000 BE: 382±13 vs 94±17; 387±47 vs 88±18; P<0.01, 1 mo for example; P<0.01). Adjacent sections immunostained with antibodies against CGRP and 5HT proved the coexistence of CGRP and 5HT in neuroendocrine cells. The data of laser scanning confocal microscopy showed that the fluorescence intensity became strong with the development of fetuses and weak in postnatal rat lungs. CONCLUSION: CGRP and 5HT positive cells changed with the development of the lung and this result suggested that these cells played an important role in the development, differentiation and maturity of the lung. The rapid changes in number during the period of birth may relate with the sharp change in function of the lung. The result of adjacent sections method proved the coexistence of CGRP and 5HT in neuroendocrine cells. This finding suggests the possibility of functional interaction of CGRP and 5HT. ......
