    Study on apoptosis and expression of HSP70 and bax in placental tissues of serious pregnancy induced hypertension

    【Abstract】AIM: To study the relationship between the apoptosis and expression of heat shock protein 70 (HSP70) and bax in placentas of serious pregnancy induced hypertension (SPIH). METHODS: Placental samples were obtained from 25 uncomplicated thirdtrimester pregnancies and from 25 cases of SPIH. Light microscopy and electron microscopy were used to identify and quantify apoptosis. HSP70 and bax were detected by immunohistochemical staining. The expression of HSP70 protein was assessed by Western blot. RESULTS: Apoptosis has been demonstrated within human thirdtrimester placental tissues. Medians and interquartile ranges of SPIH group were 0.36%(0.20%-0.46%)and those of the normal control were 0.15% (0.09%-0.21%). The incidence of apoptosis was higher in placentas of SPIH (P<0.05). The positive rates of HSP70 were 100% in SPIH group and 93% in control, but the intensity of staining was significantly different between these two groups (P<0.01). In comparison to those in the normal control, the expression of HSP70 increased in placentas of SPIH group. The positive rates of bax were 84% in SPIH group and 43% in control, respectively (P<0.01). CONCLUSION: Increased placental apoptosis in SPIH and the upregulation of expression of bax are simultaneous. The expression of HSP70 does not increase resistance to placental apoptosis in SPIH and it may play a role in the pathophysiologic mechanism of PIH. ......
