     Immunotherapy of rat osteosarcoma using tumor vaccine from fusion of dendritic cells with the osteosarcoma cells

    【Abstract】 AIM: To evaluate the therapeutic effectiveness of tumor vaccine from fusion of dendritic cells (DCs) with the osteosarcoma cells in osteosarcoma. METHODS: The UMR106 cell line was injected subcutaneously into the SD rat thigh laterally to establish the animal model. DCs obtained from SD rat bone marrow were propagated in vitro under the condition of rGMCSF, rIL4 and nrhTNFα. DCs were purified by monoclonal antibody OX62 and magnetic beads. Then the UMR106 cell line was fused with DCs to obtain the tumor vaccine and the therapeutic effectiveness of the tumor vaccine to the original tumor and lung metastasis in the animal model was observed. RESULTS: Six weeks after the treatment with tumor vaccine, the weight of the original tumor and the rate of lung metastasis in the therapeutic group were both largely reduced compared with those in control groups (P<0.01). CONCLUSION: The fusion tumor vaccine perhaps can be used in the clinical treatment of osteosarcoma. ......
