    【Abstract】 AIM: To investigate the effects of plateletactivating factor (PAF) on the proliferation of rat airway smooth muscle cells (ASMCs) and the expression of nuclear factorκB (NFκB). METHODS: MTT assay and flow cytometry were used to analyze the effects of PAF on the proliferation of ASMCs in vitro and NFκB activity change of the ASMCs stimulated by PAF was examined by electrophoretic mobility shift assay (EMSA). The effects of Nacetylcysteine (NAC) on the function of PAF were also studied. RESULTS: PAF (1×10-6-1×10-9 mol/L) stimulated the cell proliferation and reached its maximal effect at 1×10-7 mol/L. The proliferative index (PI) of the ASMCs significantly increased(P<0.05)and the NFκB activity was enhanced obviously(P<0.01) following stimulation of PAF (10-7 mol/L). NAC (20 mmol/L) partly inhibited the cell proliferation and the increased NFκB activity induced by PAF (P<0.05). CONCLUSION: PAF can stimulate the proliferation of ASMCs, which may be achieved partly through the activation of NFκB pathway. ......
