    Killing effect of adenovirus mediated HSVtk/GCV system on glioma C6 cell

    【Abstract】 AIM: To observe killing action of HSVtk/GCV system on glioma cells. METHODS: C6 cells were transfected by adenovirus with LacZ gene, then stained with Xgal for measuring the adenovirus transfected efficiency. C6 cells survival and bystander effect were observed after adding 10 mg·L-1 GCV to transfected cells. RESULTS: The more the multiplicity of infection (MOI), the more the adenovirus transfected efficiency to C6 cells (r=0.819, P<0.05). 100% transfected efficiency needed MOI 100. The killing action of HSVtk/GCV on C6 cells, depended on the amount of AdCMVtk (r=0.870, P<0.05). There was high bystander effect. CONCLUSION: Adenovirus mediated HSVtk/GCV system has good killing effect on glioma C6 cells. ......
