     CpG oligonucleotides in mediating osteoclast differentiation

    【Abstract】 AIM: CpG oligonucleotides were studied in their mediation of osteoclast differentiation. METHODS: Mouse bone marrow mononuclear cells were separated and simple bone marrow mononuclear cells or bone marrow mononuclear cells were incubated separately with 30 μg/L MCSF, 30 μg/L MCSF and 20 μg/L RANKL or with 30 μg/L MCSF and CpG ODN of different concentrations for 1-4 d. RESULTS: CpG ODN induced bone marrow mononuclear cells to differentiate into TRAPpositive osteoclasts and these cells expressed calcitonin receptors. Northern blot showed that the expression of TNFα mRNA increased with the increase of ODN from 5 nmol/L to 100 mol/L and it also increased gradually with prolonging time. CONCLUSION: CpG ODN stimulates the transformation of osteoclast, thus CpG ODN providing a potential therapeutic approach in treating bone diseases. ......
