     Effect of janus kinase/signal transducer and activator of transcription pathway on expression of tumor necrosis factorα in liver of septic rats

    【Abstract】 AIM: The purpose of the present study was to investigate the effects of janus kinase/signal transducer and activator of transcription (JAK/STAT) pathway on the expression of tumor necrosis factorα(TNFα) in hepatic tissues of rats with sepsis. METHODS: Using a sepsis model by cecal ligation puncture (CLP), 78 male Wistar rats were randomly divided into normal control group (n=6), CLP group (n=24), AG490 treatment group (n=24) and rapamycin (RPM) treatment group (n=24). At serial time points in each group, animals were sacrificed, hepatic tissue samples were harvested to determine TNFα mRNA and protein expression levels. RESULTS: Compared with those of normal controls, the TNFα mRNA levels significantly elevated in the liver 2-48 h after CLP (P<0.05) and the protein levels significantly increased at 2-48 h, respectively (P<0.05). Both treatment with either AG490 or RPM could markedly reduce hepatic TNFα mRNA and protein expressions at 2, 6, 24 and 48 h following CLP (P<0.05). CONCLUSION: These data suggested that CLP sepsis challenge can increase TNFα mRNA and protein expressions, which may be involved in JAK/STAT pathway. Inhibiting the activation of JAK/STAT pathway can reduce hepatic TNFα mRNA and protein expressions, which may help to prevent the tissue injury after sepsis. ......
