    云南含笑花粉萌发研究

    摘 要: 应用离体培养和人工授粉的方法对云南含笑花粉的萌发进行了研究。栽培和野生云南含笑花粉的萌发率

    有很大的差异, 分别为 20%和 90%。首次报道了云南含笑的花粉粒在离体培养基上萌发 2 条花粉管的现象。云南

    含笑花粉在灰岩含笑花柱头上的萌发率和萌发时间与离体培养基上的相同, 表明灰岩含笑的柱头对云南含笑的花

    粉没有排异现象。

    Abstract : Th is paper dealsw ith the po llen germ inat ion ofM ichel ia y unnanensis in v itro and on the

    st igm a ofM ichel ia ca l icicola. The germ inat ing rate of the po llen ofM. y unnanensis in w ild f ield is

    differen t f rom that of po llen of M. y unnanensis cu lt ivated in Kunm ing Bo tan ical Garden, nam ely

    90% and 20% respect ively . The phenom enon that two po llen tubers, w h ich germ inate f rom

    po llen, is ob served fo r the f irst t im e . There, is no t reject ion betw een the st igm a ofM. ca l icicola

    and the po llen ofM. y unnanensis becau se the germ inat ing rate and t im e on the st igm a ofM ichel ia

    ca lcicola is sim ilar to that in v itro .

    云南含笑(M ichel ia y unnanensis)是中国西南地

    区的特有类群, 早春开花, 花白色, 花期长, 且具有很

    强的耐干旱、耐贫瘠的能力, 是一种观赏价值较高的

    常绿灌木, 已在园林绿化上广泛栽培。在进行木兰科

    植物的杂交育种中, 我们以云南含笑为亲本培育了

    一系列杂交种, 部分杂交种已登记了新品种权。在杂

    交育种中, 我们对云南含笑花粉的萌发进行了研究,旨在掌握其生殖生物学特性, 提高杂交育种的效率。

    1 材料与方法

    1 . 1 材料

    云南含笑的花粉采自中国科学院昆明植物园的

    栽培植株和昆明西山野生植株。将刚开的花朵采回

    实验室, 然后用镊子取下雄蕊, 置于洁净的纸上; 将

    收集的花粉干燥保存于 4℃的冰箱中备用。以灰岩

    含笑(M. ca lcicola)为杂交母本。

    1 . 2 离体培养

    采用悬滴培养法。培养基为10%蔗糖+ 0. 01%

    硼酸溶液。滴1 滴该培养液于凹形载玻片的凹处, 撒

    少量花粉于培养基上, 并混匀, 倒转玻片置于盛有水

    的培养皿上, 放入 25℃的人工气候箱中培养, 每隔

    1 h观察1 次, 记录观察结果 ......
