     Changes of hydroxyproline and oxidativestress indices in bleomycininduced pulmonary fibrosis in rats

    【Abstract】 AIM: To discuss the changes of hydroxyproline and the role of oxidationantioxidation disequilibrium in pulmonary fibrosis induced by bleomycin (BLM) in rats. METHODS: A single dose BLM was intratracheally injected to induce pulmonary fibrosis in rats, so as to observe the changes of tissue hydroxyproline and oxidativestress markers during the process. The animals were killed on the 3rd, 7th, 14th and 28th d separately. The pathological model was evaluated by HE and Masson dyeing method, and the hydroxyproline of lung tissue was determined. At the same time, malonaldehyde (MDA) content, superoxide dismutase (SOD) activity, reduced glutathione/oxidized glutathione (GSH/GSSG) ratio of lung tissue were also detected. RESULTS: From 7 d to 28 d, the hydroxyproline of lung tissue was increased in BLM groups compared with that in control groups, especially on the 28th d (P<0.05). On the 7 d and14 d, the tissue MDA were enhanced while the activity of SOD was decreased compared with those of control (P<0.05). The GSH/GSSG ratio was lower than that of control on the 3 d and 7 d (P<0.05). CONCLUSION: Tissue hydroxyproline can be a marker of BLMinduced pulmonary fibrosis. The oxidationantioxidation disequilibrium of cells and matrix may play a role in the pathological process. ......
