     Effect of TNFα on expression of human glucocorticoid receptors

    【Abstract】 AIM: To study the effect of TNFα on the expression of human glucocorticoid receptor (GR) so as to explore the relationship of inflammatory cytokines and glucocorticoid resistance. METHODS: Human peripheral blood mononuclear cells (PBMCs) were isolated and the mRNA expression of GRα and GRβ in PBMC after the culture with TNFα or without TNFα was detected by RTPCR. RESULTS: No significant difference was found between the expression of GRα mRNA in PBMCs with a 24hour TNFα stimulation and that without TNFα stimulation (P>0.05) and GRα/GAPDH mRNA was 1.16±0.73 and 1.20±0.51 respectively. But the expression of hGRβ mRNA in PBMCs with a 24hour TNFα stimulation was significantly higher than that without TNFα stimulation (P<0.05). GRβ/GAPDH mRNA was 1.62±0.59 and 0.81±0.66 respectively. CONCLUSION: TNFα can increase the expression of glucocorticiod receptor β, which may be one of the mechanisms of glucocorticoid resistance induced by inflammatory cytokine. ......
