Correlation of interleukinª²12 and severity of acute necrotizing pancreatitis in rats

    ¡¾Abstract¡¿ AIM: To investigate the change of serum interleukinª²12(ILª²12) concentration in acute necrotizing pancreatitis (ANP) and its correlation with the severity of pancreas damage. METHODS: Eightyª²eight Wister rats were randomly divided into three groups. ANP were induced in Group ANP (Group A, n=32) and Group ILª²10 (Group B, n=32) by injecting intraperitoneally three times with 60 g/L Lª²Arginine (3¡Á1 mg/g) at an interval of 1 h. Group control (Group C, n=24) only received saline intraperitoneal injection. Group B was treated with 10 000 unites of intraperitioneal recombinment human ILª²10 at 2nd, 5th and 8th h after the Lª²arginine injection. The rats were killed at 6th, 12th, 24th and 36th h after the last Lª²arginine injection. Pancreas histological characteristics were scored by light microscopy and serum amylase, ILª²12 and ILª²6 concentrations were detected using enzyme substrate assay and ELISA method. RESULTS: The pancreas histological scores, serum amylase, ILª²12 and ILª²6 concentrations in Group A were markedly higher than those in Group C (P<0.05). After the administration of ILª²10, the pancreas histological scores, serum amylase, ILª²12 and ILª²6 concentrations in Group B markedly decreased (P<0.05). Positive correlation was observed between ILª²12 value and pancreas histological scores (r= 0.618, P<0.001) and serum amylase concentration (r=0.321, P<0.005). CONCLUSION: The serum concentrations of ILª²12 and ILª²6 markedly increase in experimental ANP. The pancreatic damage is alleviated after the administration of ILª²10. Positive correlation is found between serum ILª²12 concentration and the severity of pancreatic damage. ......
