Transformation of Streptococcus pneumoniae in vivo by green fluorescence protein

    ¡¾Abstract¡¿ AIM: To study when and where streptococcus pneumoniae (S.pn) forms transformation in vivo so as to study the influence of pneumococcal transformation on its virulence. METHODS: S.pn 22G whose transformation forming could be reported by enhanced green fluorescence protein (EGFP) was constructed and was identified by restricted enzyme digestion, transformation experiment, RTª²PCR and DNA sequencing. Fluorescence was observed by fleorescence microscope. RESULTS: The fluorescence emitted by 22G which was isolated from BABL/c mice or cultured in vitro was observed. CONCLUSION: S.pn can form transformation in vivo and using EGFP as report protein, pneumococcal transformation forming can be detected in vivo.
