【Abstract】 AIM: To study when and where streptococcus pneumoniae (S.pn) forms transformation in vivo so as to study the influence of pneumococcal transformation on its virulence. METHODS: S.pn 22G whose transformation forming could be reported by enhanced green fluorescence protein (EGFP) was constructed and was identified by restricted enzyme digestion, transformation experiment, RTPCR and DNA sequencing. Fluorescence was observed by fleorescence microscope. RESULTS: The fluorescence emitted by 22G which was isolated from BABL/c mice or cultured in vitro was observed. CONCLUSION: S.pn can form transformation in vivo and using EGFP as report protein, pneumococcal transformation forming can be detected in vivo.
