Establishment and biological characters of cell line GCTª²0404 from human giant cell tumor of bone

    ¡¾Abstract¡¿ AIM: To establish a human giant cell tumor (GCT) cell line of bone. METHODS: The specimens identified as GCT by pathological diagnosis were cultured primarily in vitro. The surviving cells underwent morphological observation, histochemical and immunohistochemical staining, cell cycling analysis, karyotype analysis and transplantation in nude mice. RESULTS: The newly established fibroblastª²like stromal cell line GCTª²0404 from human giant cell tumor of bone was continuously cultured 100 passages in approximately 1 year and shared the features of the primary tissue in morophology, histochemistry and immunohistochemistry. The doubling time of the cells was about 39.7 h. The cell cycle distribution analysis showed: G1 67.5%, G2 8.9% and S 23.6%. The cells were not contaminated by mycoplasma and the tumor formation rate was 100% after transplantation in nude mice. CONCLUSION: The GCTª²0404 is a cell line derived from human giant cell tumor of bone and can be used as a model for further studies. ......
