2450MHz微波对人肝癌细胞p27mRNA表达的影响

    摘 要:目的 观察2450MHz微波对人肝癌细胞p27mRNA表达的影响,以探讨其抑制肝癌细胞存活的分子机制. 方法 转染p27基因的人肝癌细胞按微波辐照强度不同分为对照组(未辐照组)、10mW·cm-2 ,20mW·cm-2 和30mW·cm-2 组,分别于微波屏蔽室内辐照1h后,用四唑盐比色试验(MTT)检测微波对肝癌细胞的抑制作用;同时用RT-PCR方法检测肝癌细胞中p27mRNA的表达,并进行灰度扫描后计算RNA的表达指数. 结果 微波辐照1h对肝癌细胞存活有明显的抑制作用,且呈剂量-效应关系.微波可以上调肝癌细胞中p27mRNA的表达,且与微波辐照强度有剂量-效应关系. 结论 微波可能通过上调p27mRNA的表达,从而抑制肝癌细胞的存活.
