关键词: myc基因;反义核酸;大鼠;平滑肌摘 要:目的 观察反义c-myc寡核苷酸对大鼠气道平滑肌增殖的抑制作用. 方法 大鼠气道平滑肌细胞原代培养,4~12代用于实验.利用Lipofectin将正义、反义及错配c-myc寡核苷酸导入大鼠气道平滑肌细胞,MTT法检测不同寡核苷酸对大鼠气道平滑肌细胞增殖的抑制作用,RT-PCR检测c-myc mRNA的表达,免疫组织化学染色检测c-Myc蛋白的表达. 结果 反义c-myc寡核苷酸可抑制大鼠气道平滑肌细胞增殖,且这种抑制作用呈浓度依赖性;反义c-myc寡核苷酸可降低c-myc mRNA的表达,同时降低了c-myc mRNA的翻译. 结论 反义c-myc寡核苷酸可抑制大鼠气道平滑肌的增殖.

Abstract:AIM To observe the antiproliferative effect of c-myc antisense oligonucleotide on rat airway smooth muscle cells.METHODS Rat airway smooth muscle cells were pri-mary cultured.4~12passages were used in experiments.Lipofectin was used to transfect antisense oligonucleotide,sense oligonucleotide and mismatched oligonucleotide for c-myc to rat airway smooth muscle cells.The antiproliferative effect was assayed by way of MTT.RT-PCR was used to de-tect the expression of c-myc mRNA.c-Myc was observed by immunohistochemistry staining.RESULTS Antisense oligonucleotide for c-myc could inhibit the proliferation of rat airway smooth muscle cells ......
