关键词: 汉坦病毒;肾综合征出血热;空斑实验;基因重组;聚合酶链式反应摘 要:目的 探明汉坦病毒汉滩型与希望山型之间能否发生基因重排,以及发生重排的频率和特点. 方法 用汉坦病毒汉滩型76-118株与希望山型PHV株混合感染Vero E6细胞,空斑形成实验挑取子代病毒克隆株,传代培养后用RT-PCR方法鉴定子代病毒基因型. 结果 子代病毒14/36来源于PHV株;12/36来源于76-118株;5/36汉滩型与希望山型分型引物扩增均为阳性;1株子代病毒L,S片断均来源于76-118株,而M片段两种引物扩增均为阳性;2株子代病毒L片断来源于76-118株,S片断来源于PHV株,M片段两种引物扩增均为阳性.1株子代病毒L,S片断均来源于76-118株,而M片段来源于PHV株. 结论 汉坦病毒汉滩型与希望山型之间可以发生基因片段的重排.
