     【摘要】 目的 探讨绿茶提取物表没食子儿茶素没食子酸酯(epigallocatechin-3-gallate,EGCG)对人胃癌MGC-803细胞株增殖的影响及其分子机制。 方法 采用MTT法测定EGCG对胃癌细胞增殖的抑制作用;运用免疫细胞化学和免疫印迹技术分别检测c-Myc和磷酸化Erk1/2蛋白表达改变。 结果 EGCG对MGC-803细胞增殖具有抑制作用,呈浓度依赖性。EGCG处理后免疫细胞化学检测c-Myc蛋白的表达下调,免疫印迹检测磷酸化Erk1/2蛋白表达下调。 结论 EGCG抑制MGC-803细胞增殖,呈浓度依赖性,其抑制肿瘤细胞增殖的效应与抑制Erk1/2蛋白磷酸化及下调c-Myc蛋白有关。

    Role of Erk1/2 pathway in the inhibitory effects of EGCG on proliferation of MGC-803 cell

    【Abstract】 Objective To probe the effects of green tea extract epigallocatechin-3-gallate(EGCG) on proliferation of human gastric cancer cell line MGC-803 and its molecular mechanisms. Methods MTT assay was used to test the inhibitory effects of EGCG on proliferation in MGC-803 cells. Expression of c-Myc in MGC-803 cells was detected by immunocytochemistry.The expression of phosphorated Erk1/2 protein in MGC-803 cells was examined using Western blot analysis.Results MTT assay has shown that EGCG significantly inhibited proliferation of MGC-803 cells in a concentration dependent manner.Immunocytochemistry detected the expression down regulation of c-Myc protein analysis treated with EGCG for 48 hours in MGC-803 cells.Western blot analysis confirmed the expression of phosphorylated Erk1/2 protein significantly down regulated.Conclusion EGCG inhibits proliferation of MGC-803 cells in a concentration dependent manner and its mechanism associated with the down regulation of c-Myc and phosphorylated Erk1/2 proteins expression. ......
