     Electrophysiology of Irbesartan in guinea pig ventricular myocytes

    【Abstract】 AIM: To investigate the effects of Irbesartan on ICaL, IK1, INa and transmembrane action potentials in guinea pig ventricular myocytes. METHODS: Membrane currents were recorded with patch clamp technique in a wholecell clamp configuration. RESULTS: ① Irbesartan (10,100,1000 nmol/L) reduced the ICaL density (PA/PF) from (-98±11) (control) to (-67±09), (-42±08) and (-21±0.4) (n=5, P<005 or 001) respectively in a concentrationdependent manner. ② Irbesartan upshifted the currentvoltage (IV) curve, but the characteristics of IV relationship were not significantly altered by Irbesartan. ③ Irbesartan blocked ICaL in a usedependent manner. ④ Steadystate activation curve of ICaL was not affected markedly, but Irbesartan (100 nmol/L) markedly shifted the steadystate inactivation curve of ICaL to the left, and accelerated the voltagedependent inactivation of calcium current, V1/2 value was (-30±5) mV in the control and (-39±7) mV in the presence of Irbesartan, the κ value was (82±07) and (116±13) respectively (n=5 ......
