中药天年饮对衰老大鼠学习记忆及脑单胺类神经递质含量的影响

     摘要:目的观察中药天年饮(Tiannianyin,TNY)对D半乳糖衰老大鼠学习记忆及脑单胺类神经递质去甲肾上腺素(NE)、多巴胺(DA)、5羟色胺(5HT)含量的影响。方法选用成年雄性SD大鼠40只,随机分为4组,每组10只:正常组、衰老模型组、TNY用药组、阴性对照组。D半乳糖连续腹腔注射制作亚急性衰老的大鼠模型,采用Morris水迷宫检测各组大鼠的空间学习记忆能力及采用高效液相色谱电化学法检测各组大鼠下丘脑、海马NE,DA,5HT的含量。结果D半乳糖衰老大鼠空间学习记忆能力明显下降并且衰老大鼠下丘脑,海马NE,DA,5HT的含量明显降低(与正常大鼠相比P<0.01);TNY可明显改善衰老大鼠的空间学习记忆能力及提高脑单胺类神经递质的含量(P<0.01或P<0.05)。结论TNY可有效调整中枢神经递质的合成及提高学习记忆能力,具有良好的延缓衰老的作用。

    Influence of Chinese Decoction TNY on Learning and Memory and Monoamine Neurotransmitter in Aging Rats Brain

    Abstract:ObjectiveTo observe the influence of Chinese decoction Tiannianyin (TNY) on learning and memory and monoamine neurotransmitter contents in Dgal aging rats brain. Methods40 adult male SD rats were used in this study and divided into 4 groups at random: normal group, aging model group, TNY treatment group, and negative control group, each group had 10 rats. The subacutely aging rats were made by injecting Dgal into the rats' abdominal cavity continuosly. The learning and memory ability of rats in each group were detected with Morris water maze. The monoamine transmitter contents in hypothalamus and hippocampus were detected with HPLCECD.ResultsThe learning and memory ability of aging rats dropped significantly compared with normal rats(P<0.01). The NE, DA, 5HT contents in hippocampus and hypothalamus of aging rats decreased significantly compared with normal rats (P<0.01).
