Curcumin regulates cyclooxygenaseª²2 activity in trinitrobenzene sulfonic acidª²induced colitis

    ¡¾Abstract¡¿ AIM: To explore the effects of nuclear factorª²¦ÊB (NFª²¦ÊB) inhibitor curcumin on the cyclooxygenaseª²2 activity in the colonic mucosa of rats with trinitrobenzene sulfonic acid (TNBS)ª²induced colitis so as to screen out an antiª²inflammatory target of curcumin.METHODS: Rats with TNBSª²induced colitis were treated with 20 g/L curcumin in the diet.Positive control rats were treated with 5 g/L sulfasalazine (SASP) and 2 g/L Nª²acetylcysteine (NAC).Negative control rats and model rats were treated with general diet.The changes in colonic mucosal histological scores were evaluated and the cell nuclear NFª²¦ÊB activity in the colonic mucosa was evaluated by electrophoretic mobility shift assay.Cytoplasmic I¦ÊB protein and cyclooxygenaseª²2 protein in colonic mucosa were detected by Western Blot analysis.Cyclooxygenaseª²2 messenger expression in colonic tissue was assessed by semiquantitative reverseª² transcription polymerase chain reaction.RESULTS: Degradations of cytoplasmic I¦ÊB protein in colonic mucosa were blocked by curcumin treatment.NFª²¦ÊB activation in colonic mucosa was suppressed in the curcuminª²treated groups.Cyclooxygenaseª²2 activity in colonic mucosa reduced in the curcuminª²treated groups.Treatment with curcumin ameliorated the histopathologic signs of rats with TNBSª²induced intestinal inflammation.CONCLUSION: NFª²kappa B inhibitor curcumin can prevent and ameliorate experimental colitis in murine model of inflammatory bowel disease (IBD) and regulate the cyclooxygenaseª²2 activity.Cyclooxygenaseª²2 can be a medicine target for IBD and curcumin is a potential medicine in treating the patients with IBD. ......
