【Abstract】 AIM: To explore the effects of nuclear factorκB (NFκB) inhibitor curcumin on the cyclooxygenase2 activity in the colonic mucosa of rats with trinitrobenzene sulfonic acid (TNBS)induced colitis so as to screen out an antiinflammatory target of curcumin.METHODS: Rats with TNBSinduced colitis were treated with 20 g/L curcumin in the diet.Positive control rats were treated with 5 g/L sulfasalazine (SASP) and 2 g/L Nacetylcysteine (NAC).Negative control rats and model rats were treated with general diet.The changes in colonic mucosal histological scores were evaluated and the cell nuclear NFκB activity in the colonic mucosa was evaluated by electrophoretic mobility shift assay.Cytoplasmic IκB protein and cyclooxygenase2 protein in colonic mucosa were detected by Western Blot analysis.Cyclooxygenase2 messenger expression in colonic tissue was assessed by semiquantitative reverse transcription polymerase chain reaction.RESULTS: Degradations of cytoplasmic IκB protein in colonic mucosa were blocked by curcumin treatment.NFκB activation in colonic mucosa was suppressed in the curcumintreated groups.Cyclooxygenase2 activity in colonic mucosa reduced in the curcumintreated groups.Treatment with curcumin ameliorated the histopathologic signs of rats with TNBSinduced intestinal inflammation.CONCLUSION: NFkappa B inhibitor curcumin can prevent and ameliorate experimental colitis in murine model of inflammatory bowel disease (IBD) and regulate the cyclooxygenase2 activity.Cyclooxygenase2 can be a medicine target for IBD and curcumin is a potential medicine in treating the patients with IBD. ......
