     Relationship between hypoxicendurance and erythropoietin in ovarian carcinoma cell line

    【Abstract】 AIM: To investigate the endurance of ovarian carcinoma cell line to hypoxia by measuring the expression of erythropoietin (EPO) and to study the role of EPO in the antiapoptosis and the genesis and development of ovarian carcinoma. METHODS: The 3AO cells plated in culture bottles were placed in an anaerobic system consisting of 950 mL/L N2 and 50 mL/L CO2 for different time periods. The cell growth was observed by MTT assay, the apoptosis rate was detected by FCM and the mRNAs expression of EPO was detected by in situ hybridization. RESULTS: Apoptosis of cells cultured under hypoxic condition increased after 6 h of hypoxic exposure compared with that at 0 h (P<0.01) and the activity of cell growth decreased obviously. The intensity of expression of EPOmRNA increased from 2 h after hypoxic exposure. The number of apoptosis cells reduced after 16 h of hypoxic exposure and the activity of cell growth gradually resumed. After 24 h of hypoxic culture, apoptosis was the same as that at 0 h. CONCLUSION: EPO is correlated with hypoxicendurance and it may be involved in antiapoptosis of ovarian cancer cells under hypoxic condition. ......
