     Expression of NFκB in rat hepatic stellate cells stimulated by acetaldehyde

    【Abstract】 AIM: To investigate the effects of acetaldehyde on the proliferation and the expression of NFκB and IκB in rat hepatic stellate cells (HSCs) and to explore its mechanism.METHODS: HSCs were isolated by means of density gradient centrifugation after digestion in pronase E and collagenase Ⅳ and the relationship between acetaldehyde and the proliferation and the expression of NFκB and IκB in HSCs were studied by MTT,immunohistochemistry and EMSA.RESULTS: ① HSCs were isolated successfully.② The freshly isolated HSCs proliferated significantly after 24 h stimulation with acetaldehyde at a concentration of 200 μmol/L [(221±010) vs (052±005),P<001].③ The expression of IκB was inhibited when it is stimulated by acetaldehyde at a concentration of 200 μmol/L [(032±002) vs (160±032),P<001].④ Thirty minutes after the stimulation of acetaldehyde (200 μmol/L),the expression of NFκB in HSC increased,and 120 min after the stimulation of acetaldehyde (200 μmol/L),the NFκB activity in HSCs reached the peak [(392±082) vs (082±004),P<001].CONCLUSION: Acetaldehyde can inhibit the expression of IκB and activate the expression of NFκB in quiescent HSCs,by both of which the proliferation of HSCs is activated. ......
