HPLC柱前衍生化法测定阿波莫斯注射液的含量

     摘要采用高效液相色谱邻苯二甲醛柱前衍生化法，测定进口药品阿波莫斯注射液中天门冬鸟氨酸的含量。以Hypersil ODS(125 mm×4 mm，5 μm)为色谱柱，20 mmol/L NaAc+φ=0.5%THF+φ=0.02%TEA(pH7.2)混合一定比例的100 mmol/L醋酸钠乙睛甲醇(100∶175∶225)作流动相，梯度洗脱。结果在0.20～1.20 μg范围内，积分面积与样品量呈良好的线性关系，r=09991，平均加样回收率为100.3%，RSD=1.8%(n=9)。

    Determination of HepaMerz Injection by Precolumn Derivatization of HPLC

    Abstract A precolumn derivatization with OPA of HPLC method was established for the determination of HepaMerz injection. Reversedphase C18column (Hypersil ODS 125 nm×4 nm,5 μm) was used.Mixture of 20 mmol/L sodium acetate, 0.5% tetrahydrrofuran and 0.02% triethylamine pH 7.2 was used as mobile phase A and mixture of 100 mmol/L sodium acetateacetonitrilmethanol (100∶175∶225) as mobile phase B. The detecting wavelength was 338 nm . There was a good linear relationship in the range from 0.20 μg to 1.20 μg(r=0.9991). The average recovery was 100.3%(n=9,RSD=1.8%).The method was simple,rapid and accurate. ......
