中国眼镜蛇毒蛋白酶natrahagin水解血小板膜糖蛋白 Ⅰ b和抑制血小板聚集的作用

     摘要 目的:观察从中国眼镜蛇(Naja naja atra)毒中纯化的蛋白酶natrahagin水解血小板膜糖蛋白Ⅰb(platelet membrane glycoprotein Ⅰb,GPⅠb)和抑制血小板聚集的作用。方法:通过SDS_聚丙烯酰胺凝胶电泳(SDS_PAGE)检测以及凝胶图像光密度分析系统分析natrahagin对血小板膜糖蛋白的水解作用。比浊法测定血小板聚集率。结果:Natrahagin选择性地水解GPⅠb,切除分子量约3.3×104的片段。Natrahagin(0025~0.1 mg/kg)静脉注射可显著抑制低浓度凝血酶(100 U/L)诱导的兔洗涤血小板聚集,效应呈剂量依赖性。结论:Natrahagin具有水解GPⅠb和抑制后者介导的血小板聚集的作用。

    Abstract Objective: To study the effect of natrahagin,a proteinase purified from Chinese cobra venom on platelet membrane glycoprotein Ⅰb(GPⅠb) and platelet aggregation. Methods:SDS_polyacrylamide gel electrophoresis(SDS_PAGE) and densitometric scanning were used to determined the hydrolysis of GPⅠb. The platclet aggregation was measured turbimetrically. Results:Natrahagin selectively degraded GPⅠb by cleaving a fragment with a molecular mass of about 3.3×104 Intravenous injection with natrahagin(0.025~0.1 mg/kg) significantly inhibited the aggregation of rabbit washed platelets induced by 100 U/L of thrombin in a dose_dependent manner. Conclusions:Natrahagin cleaved GPⅠb and exerted an inhibitory effect on GPⅠb_dependent platelet aggregation. ......
