【Abstract】 AIM: To screen genes encoding proteins which can interact with BRCA2 via yeast twohybrid system, confirm the interaction and analyze their functional linkage. METHODS: 3′region of BRCA2 gene was used to construct the “bait” plasmid and to screen the cDNA library of normal human mammary gland. Coimmunoprecipitation, mammalian twohybrid and Luciferase assay were used to analyze the association and putative function. RESULTS: Many genes encoding proteins interacting with BRCA2, including the FHL2, were pulled out by yeast twohybrid screening. The in vivo binding and functional relation between BRCA2 and FHL2 was demonstrated by coimmunoprecipitation, mammalian twohybrid assay and luciferase detection. CONCLUSION: The interaction and the functional linkage between BRCA2 and FHL2 was confirmed and which provides fresh insights into the biological functions of BRCA2. ......
