Interaction between BRCA2 and the LIM domain only protein FHL2

    ¡¾Abstract¡¿ AIM: To screen genes encoding proteins which can interact with BRCA2 via yeast twoª²hybrid system, confirm the interaction and analyze their functional linkage. METHODS: 3¡äregion of BRCA2 gene was used to construct the ¡°bait¡± plasmid and to screen the cDNA library of normal human mammary gland. Coª²immunoprecipitation, mammalian twoª²hybrid and Luciferase assay were used to analyze the association and putative function. RESULTS: Many genes encoding proteins interacting with BRCA2, including the FHL2, were pulled out by yeast twoª²hybrid screening. The in vivo binding and functional relation between BRCA2 and FHL2 was demonstrated by coª²immunoprecipitation, mammalian twoª²hybrid assay and luciferase detection. CONCLUSION: The interaction and the functional linkage between BRCA2 and FHL2 was confirmed and which provides fresh insights into the biological functions of BRCA2. ......
