糖尿病大鼠肾小管上皮细胞转化与肝细胞生长因子和Smad7蛋白的表达

    【摘要目的 探讨糖尿病肾病(DN)时肾小管上皮细胞转化以及肝细胞生长因子(HGF)、Smad7蛋白在其中可能发挥的作用。 方法 60只Wistar大鼠均行右肾切除术,伤口愈合后随机分为右肾切除对照组和糖尿病组。腹腔注射链脲佐菌素诱发大鼠糖尿病模型。采用免疫组化法检测角蛋白18(CK18)、α平滑肌肌动蛋白(αSMA)、HGF和Smad7蛋白的表达;流式细胞术检测αSMA和HGF的表达;蛋白质免疫印迹法检测Smad7蛋白的表达。 结果 糖尿病组较对照组CK18的表达降低,αSMA的表达上升,HGF和Smad7蛋白的表达表现为先上升后下降。 结论 糖尿病时,肾小管上皮细胞可发生表型转化,转化为肌成纤维细胞。HGF和Smad7蛋白表达下降可能与该模型中肾小管上皮细胞转化有关。

    【Abstract Objective To investigate the relationship of tubular epithelialmyofibroblast transdifferentiation and the expressions of hepatocyte growth factor (HGF) and Smad7, and to elucidate the role of HGF and Smad7 in diabetic nephropathy. Methods Diabetes was induced in male Wistar rats with right nephrectomy and streptozotocin (STZ) administration. The expressions of cytokeratin 18 (CK18), αsmooth muscle actin (αSMA), HGF and Smad7 were assayed with immunohistochemistry. The expressions of αSMA, HGF were assayed with flow cytometry. The expression of Smad7 was assessed by Western blot. Results Compared with those in kidneys of the control group, diabetic kidneys showed downregulated expression of CK18 and upregulated expression of αSMA. The expressions of HGF and Smad7 were increased significantly in the kidneys of diabetic rats at first, then they decreased gradually, but were still higher than those of control. Conclusion The tubular epithelial cells may undergo phenotypic alterations and change to myofibroblasts. The absence of upregulation of HGF and Smad7 may be related with the transdifferentiation of tubular cells in this animal model. ......
