脂质体介导人纤溶酶原k13基因诱导人肝癌细胞凋亡

     摘要:目的 观察脂质体介导人纤溶酶原k13基因诱导体外培养人肝癌细胞凋亡的效应。方法 构建人纤溶酶原k13基因真核表达载体pcDNAk13并以阳离子脂质体介导pcDNAk13转染体外培养人肝癌细胞,经Heochst 33258染色后,在荧光显微镜下观察细胞核的形态。结果 转染pcDNAk13组可见致密浓染的凋亡细胞核。结论 脂质体介导人纤溶酶原k13基因能诱导体外培养人肝癌细胞凋亡。

    Human Plasminogen k13 gene induces apoptosis in human hepatocellular carcinoma cells by polycationic liposomemediated transfection

    Abstract: Objective To investigate the effect on apoptosis of cultured human hepatocellular carcinoma (HCC) cells induced by the human plasminogen kringle 13 (k13) gene by polycationic liposomemediated transfection. Methods recombinant eukaryotic expression vectors pcDNAk13 encoding human plasminogen k13 gene were constructed and transferred to cultured HCC cells by polycationic liposomemediated gene transfer method. After stained with Hoechst 33258 staining solution, and the stained nuclei were observed under a fluorescence microscope. Results condensed bright apoptotic nuclei were readily observed among cells with transfected pcDNAk13. Conclusion human plasminogen k13 gene induces apoptosis in cultured HCC cells by polycationic liposomemediated transfection. ......
