西替伪麻碱缓释片中西替利嗪在Beagle犬体内生物利用度研究

     摘要:目的 建立高效液相色谱法测定西替伪麻缓释片中西替利嗪在Beagle犬体内血药浓度并进行生物利用度评估。方法 以二氯甲烷提取溶剂,异丙嗪为内标;采用C18柱,以乙腈0.02 mol/L的醋酸钠水溶液高氯酸三乙胺(体积比50∶50∶0.5∶0.1),用5 mol/L的氢氧化钠调节pH 5.2为流动相,检测波长230 nm。结果与结论 本文方法提取率高,回收率好,自制片的相对生物利用度93.0%。

    Bioavailability of cetirizine in cetirizinepseudoephedrine sustainedrelease tablets in beagle dogs

    (Institute of Pharmaceutical Sciences,Guangdong College of Pharmacy,Guangzhou 510006,China)Abstract: Objective To establish an HPLC assay for the determination of blood plasma concentration of cetirizine in cetirizinepseudoephedrine sustainedrelease tablets in beagle dogs.Method Cetirizine was extracted by methylene chloride. Promethazine was used as an internal standard and the separation was performed on a reversed phase Diamonsil C18 (5 μm,250 mm×4.6 mm) employing a UVdetection set at 230 nm at ambient temperature. The mobile phase consists of acetonitrile0.02 mol/L acetate water solutionperchloric acidtriethylamine (50∶50∶0.5∶0.1) adjusted to pH 5.2 with 5 mol/L NaOH. Result and Conclusion The assay is linear from 50 to 3200 ng/ml with a detection limit of 15 ng/ml. The relative bioavailability of cetirizine in the tablets was 93.0%. ......
