壳聚糖对泊洛沙姆液体栓剂基质影响的体外研究

     摘 要 目的 考察壳聚糖对以泊洛沙姆(poloxamer)为基质的液体栓剂的物理化学性质的影响,为两者的配伍使用提供试验依据。方法 将5种不同浓度壳聚糖质量分数ω=0%、0.1%、0.2%、0.3%、0.4%合并泊洛沙姆溶液[P407和P188的质量分数分别为15%和19%]作为液体栓剂的基质,测定不同质量分数壳聚糖对液体栓剂基质的胶凝温度、胶凝强度和生物黏附力的影响。结果 当壳聚糖的质量分数从0%上升至0.4%时,基质的胶凝温度依次为27.8、28.0、28.1、28.4、28.8 ℃;胶凝强度依次为12、40、86、107、168 s;生物黏附力值依次为36.9、50.5、54.7、62.2、85.6(×102dyne/cm2)。说明随着壳聚糖浓度增高,泊洛沙姆液体栓剂基质的胶凝温度、胶凝强度和生物黏附力均逐渐升高。结论 壳聚糖可改善以泊洛沙姆为基质的液体栓剂的物理化学性质,值得进一步研究。

    Effects of chitosan on the liquid suppository composed of poloxamers in vitro

    Abstract Objective To study the effects of different concentrations of chitosan on the liquid suppository composed of poloxamers in vitro. Methods Five different concentrations of chitosan (0%, 0.1%, 0.2%, 0.3% and 0.4%, W/W) and poloxamers (P407∶P188=15%∶19%, W/W) were used as the liquid suppository bases. The changes of the bases were determined by measuring gel temperature, gel strength and bioadhesive force.
