血小板活化因子对嗜酸性粒细胞活化作用的实验研究

     摘 要 目的 探讨血小板活化因子(PAF)在低密度嗜酸性粒细胞(HE)的产生和嗜酸性粒细胞(EOS)脱颗粒的过程中所起的作用以及二者之间的关系,以了解HE的产生机制。方法 分离正常人外周血EOS,测定PAF对EOS脱颗粒、EOS密度的作用。 结果 PAF能刺激EOS脱颗粒,其最大作用浓度为10-9 mmol/L,即可使29.97%的正常密度EOS(NE)转变为HE,二者之间呈显著正相关。结论 EOS脱颗粒是HE产生的原因之一;PAF可以刺激EOS脱颗粒使NE转变为HE,PAF刺激EOS脱颗粒可能是支气管哮喘等过敏性疾病体液中HE增高的原因。

    Experimental study on effect of plateletactivating factor on eosinophil activation

    Abstract Objectives this study was performed to elucidate the effects of plateletactivating factor (PAF) on eosinophil activation. Methods separated eosinophils(EOS) from the peripheral blood of normal subjects, and measured the effects of PAF on eosinophil activation. Results PAF induced the release of eosinophil peroxidase (EPO), and the generation of hypodense eosinopil(HE). There was significant correlation between PAFinduced EPO release and HE generation. Conclusion Eosinophil degranulation is one of the reasons for which HE develops; PAF can induce EOS degranulation, so that generate HE. PAF may be responsible for the raised HE count in eosinophiliaassociated diseases(e.g., asthma). ......
