高效液相色谱法测定大鼠血浆中游离脂肪酸浓度

     摘 要 目的 建立大鼠血浆中游离脂肪酸(FFAs)浓度的测定方法。方法 以十七烷酸为内标,用ψ(异丙醇∶正庚烷∶2 mol/L磷酸)=(40∶10∶1)提取,溴苯乙酮衍生化,流动相为ψ(乙腈∶水)=(81∶19),流速1.0 mL/min,柱温55 ℃,分析柱Novapak C18(250 mm×4.6 mm,4 μ m),紫外检测波长242 nm。结果 各FFAs最低检测限为0.6 μmol/L,平均回收率为(100.1±20)%,批内、批外RSD<6.5%。结论 本法稳定、灵敏、可靠,适用于大鼠血浆中FFAs浓度测定。

    Determination of free fatty acids in rat plasma by HPLC

    Abstract Objective To develop a HPLC method for determination of free fat acids(FFAs) in rat plasma. Methods Using heptadecanoic acid as the internal standard, FFAs were extracted from rat plasma with a mixture of isopropanol∶heptane∶ phosphoric acid (2 mol/L)=40∶10∶1. FFAs were derived by phenacyl bromide. The mobile phase was acetonitrile∶water(81∶19) at 1.0 mL/min with UV detection at 242 nm and column temperature was 55 ℃. The analytic column was NovaPAK C18(250 mm×4.6 mm, 4 μm). Results The limit of detection was 0.6 μmol/L. The average recovery was (100.1±2.0)%, and the relative standard deviation of intraday and interday were limited within 6.5%, respectively. Conclusion The method was stable, sensitive and accurate, and suitable for determination of FFAs in rat plasma. ......
