一氧化碳对急性肺损伤时炎性细胞因子作用的初步研究

     【摘要】 目的 探讨外源性一氧化碳(CO)在急性肺损伤(ALI)发病中产生的影响及其可能的机制。方法 随机将30只条件相当的SD大鼠分为三组:健康对照组、LPS组及LPS+CO组。24h测动脉血和支气管肺泡灌洗液(BALF)中细胞因子IL-8、IL-2及TNF含量。并取三组大鼠肺组织,做病理切片及电镜片进行比较。结果 LPS组,在静脉注射LPS(大肠杆菌内毒素)后24h其动脉血和BALF中细胞因子IL-8、IL-2及TNF含量明显增加,显著高于正常对照组,但是同时给予CO吸入的LPS+CO组,细胞因子IL-8、IL-2及TNF含量虽有增高,但与正常对照组之间差异无显著性,而与LPS组差异有显著性。病理切片及电镜片的比较亦提示LPS+CO组其炎性改变较LPS组轻。结论 给大鼠静脉注射LPS造成ALI的同时给予低浓度CO气体吸入,可降低细胞因子水平从而减轻急性肺损伤的炎性反应。

    The study of carbon monoxide from outside effect with cytokine in acute lung injury

    【Abstract】 Objective To explore the possible involvement of carbon monoxide (CO) in the development of lipopolysaccharide (LPS)-induced acute lung injury (ALI). Methods We build three groups: control rats, LPS rats, LPS+CO rats. We determined the concentration of cytokine, include interleukin-8(IL-8),interleukin-2(IL-2) and tumor necrosis factor (TNF) in carotid artery and in bronchoalveolar lavage fluid (BALF)from male SD rats 24h after injection with LPS via caudal vein. Then we got pathological section and electron microscope section of these rats’ lung.Results The cytokine of carotid artery and BALF was significantly higher in the rats at 24h after LPS injection than in control rats P<0.01. The level of cytokine in CO+LPS rats was also little higher than in control rats ......
