葛根素对血管性痴呆大鼠海马中低氧诱导因子1α和红细胞生成素表达的影响

    摘要:目的 观察葛根素对血管性痴呆(VD)大鼠海马中低氧诱导因子1α和红细胞生成素表达的影响并探讨其可能机制。方法 采用大鼠双侧颈总动脉永久性阻断法(2VO)建立VD的动物模型,SD大鼠随机分为假手术组、2VO组和葛根素组,每组又分为1周、3周、6周、2月(n=6)四个时间点;应用Y型水迷宫实验测定动物的学习记忆能力,采用免疫组化法检测大鼠海马CA1区HIF1α和EPO蛋白的表达。结果 ①2VO组和葛根素组大鼠游全程时间和误入盲端次数均显著增加,但各时间点葛根素组大鼠的学习记忆成绩均显著优于2VO组。②各时间点,2VO组和葛根素组大鼠海马CA1区HIF1α和EPO的表达均较假手术组明显增高,但葛根素组两种蛋白的表达均较2VO组明显降低。结论 葛根素可提高缺血脑组织细胞内氧浓度,改善学习记忆功能,对VD具有确切的保护作用。

    Effects of puerarin on the expression of hypoxia inducible factor1α

    ABSTRACT: Objective To observe the effects of puerarin on the expression of hypoxia inducible factor1αand erythropoietin in the hippocampus of vascular dementia rats and to investigate its possible mechanisms. Methods Rats of experimental group were treated with a permanent bilateral common carotid arteries (CCA) occlusion (2VO) to establish vascular dementia model. Male SD rats were randomly divided into three groups: shamoperated group, 2VO group and puerarin group(twentyfour rats in each group). Rats were evaluated on learningmemory ability by Ytype water maze test. At corresponding time points at 1w, 3w, 6w and 2mon, the dynamic expression of HIF1α and EPO in CA1 region of hippocampus was determined by immunohistochemical method. Results ① The swimming time and error times of rats in 2VO group and puerarin group were significantly increased; but at all time points the rats in puerarin group showed better performance than those in 2VO group. ② Both in 2VO group and puerarin group, the number of HIF1α and EPO positive cells in hippocampal CA1 region were much more than that in shamoperated group; but the expression of HIF1α and EPO at each point in puerarin group was distinctly less than that in 2VO group. Conclusion Puerarin can increase the intracellular oxygen concentration and improve learningmemory ability during cerebral ischemia, and it has definitely protective effects on VD. ......
