作者简介:吴海琴(1960),女(汉族),教授,硕士研究生导师,主要从事脑血管疾病的研究.摘要:目的 观察葛根素对血管性痴呆(VD)大鼠海马中低氧诱导因子1α和红细胞生成素表达的影响并探讨其可能机制。方法 采用大鼠双侧颈总动脉永久性阻断法(2VO)建立VD的动物模型,SD大鼠随机分为假手术组、2VO组和葛根素组,每组又分为1周、3周、6周、2月(n=6)四个时间点;应用Y型水迷宫实验测定动物的学习记忆能力,采用免疫组化法检测大鼠海马CA1区HIF1α和EPO蛋白的表达。结果 ①2VO组和葛根素组大鼠游全程时间和误入盲端次数均显著增加,但各时间点葛根素组大鼠的学习记忆成绩均显著优于2VO组。②各时间点,2VO组和葛根素组大鼠海马CA1区HIF1α和EPO的表达均较假手术组明显增高,但葛根素组两种蛋白的表达均较2VO组明显降低。结论 葛根素可提高缺血脑组织细胞内氧浓度,改善学习记忆功能,对VD具有确切的保护作用。

ABSTRACT: Objective To observe the effects of puerarin on the expression of hypoxia inducible factor1αand erythropoietin in the hippocampus of vascular dementia rats and to investigate its possible mechanisms. Methods Rats of experimental group were treated with a permanent bilateral common carotid arteries (CCA) occlusion (2VO) to establish vascular dementia model. Male SD rats were randomly divided into three groups: shamoperated group, 2VO group and puerarin group(twentyfour rats in each group). Rats were evaluated on learningmemory ability by Ytype water maze test. At corresponding time points at 1w, 3w, 6w and 2mon, the dynamic expression of HIF1α and EPO in CA1 region of hippocampus was determined by immunohistochemical method. Results ① The swimming time and error times of rats in 2VO group and puerarin group were significantly increased; but at all time points the rats in puerarin group showed better performance than those in 2VO group. ② Both in 2VO group and puerarin group, the number of HIF1α and EPO positive cells in hippocampal CA1 region were much more than that in shamoperated group; but the expression of HIF1α and EPO at each point in puerarin group was distinctly less than that in 2VO group. Conclusion Puerarin can increase the intracellular oxygen concentration and improve learningmemory ability during cerebral ischemia, and it has definitely protective effects on VD. ......
